Help: One of the colors in my layers screwed up after saving the file.

Currently working with Sketchbook Pro 4. I was giving highlights to my drawing with a layer of itself, I go off to do something so I save and close the window. When I come back, I open up the drawing and the highlight’s layer is all messed up. The colors changed to grayish scales in some places and it just looks awful. I have no clue how to fix this and it is starting to really frustrate me. The files is saved as TIFF.
